TitleThe effect of xenon light toxicity on the RPE cells: an in vitro study
PurposeTo compare the phototoxic effect of xenon light in vitrectomy using an A2E-laden retinal pigment epithelial (RPE) model.
MethodsIn vitro, a monolayer of A2E-laden RPE cells was exposed to xenon light coming from 20 gauge fibers positioned at 1cm above the cells. A comparison was made between light sources from DORC and ALCON.
Variables for both light sources were the time of exposure (15, 30 or 60 minutes) and the intensity of light (100, 50, 35 or 20%). The wavelength was also a variable for the DORC light source (420-435-475-515nm), whereas the ALCON light source contained a mixture of wavelengths. The cell viability was assessed with a Live Dead Kit® and analyzed with confocal microscopy.
ResultsThe viability of the cells was influenced by all 3 parameters: the intensity, time period and the wavelength. The shorter the wavelength (435nm/420nm), the more cells died. Additionally, a nice dose response between the intensity of light and toxicity was observed; with an intensity of 100% the mortality was significantly increased to more than 50% for both light sources. A combination of a long time period (60 min), a high intensity (100%) for the ALCON light source and a short wavelength (420nm/435nm) for the DORC light source resulted in even more cell death. When the operating room settings were mimicked (intensity 20 to 35%, duration of 60 min for the ALCON light source; and intensity 20 to 35%, duration of 60 min, wavelength of 475nm for the DORC light source), both light sources were shown to be safe for the RPE cells, with a viability of about 80% and 70%, respectively.
ConclusionBoth light sources were equally safe when clinically relevant time period, intensity and wave length were used.
